Inhibition of keto acid oxidation by plague toxin.

چکیده
The major objective of this work has been to learn the effect of plague toxin on several enzymatic systems in vitro. The availability of purified preparations of plague toxin (Ajl et al., 1955) permitted us to carry out these investigations. In this report, experiments are described in which purified plague toxin has been added to actively respiring cell-free extracts and crude mouse liver homogenates. It has been found that the toxin inhibits the oxidation of a-keto acids by these enzymatic preparations.

Benign prostatic hyperplasia (BPH) is a non-malignant enlargement of the prostate gland. Itis a leading disorder of the elderly male population. Excessive production of dihydrotestosteronehas been implicated in this pathological condition. Steroidal 5α-reductase is a membrane boundNADPH dependent enzyme which is responsible for the conversion of testosterone (T) todihydrotestosterone (DHT). The...

shown to actively utilize branched chaini amino acids and to produce alanine. The coniversioni of [1-"C]leucine to 14CO2 or to the [1-14C]keto-acid of leucine (a-keto-isocaproate) was linear for at least 2 h of incubation; the production of "CO2 from [1-'4C]leucine was saturable with a Km = 6.3 mM and a maximiium oxidation rate (Vmax) = 31 nmiol/mg protein per 120 min. Clofibric acid selectivel...
متن کاملPeptide and keto acid utilization in replacing phenylalanine in adaptive enzyme synthesis.
Glycylasparagine has been found to be more effective than aspartic acid in reversing the inhibition of adaptive enzyme synthesis by cysteic acid or P-hydroxyaspartic acid or combinations of these synergistic inhibitors (1). The results were interpreted to indicate a pathway of utilization of the peptide not involving the inhibited enzymatic utilization of the free amino acid. Also, histidine pe...
متن کاملBeneficial effects of trimetazidine in ex vivo working ischemic hearts are due to a stimulation of glucose oxidation secondary to inhibition of long-chain 3-ketoacyl coenzyme a thiolase.
High rates of fatty acid oxidation in the heart and subsequent inhibition of glucose oxidation contributes to the severity of myocardial ischemia. These adverse effects of fatty acids can be overcome by stimulating glucose oxidation, either directly or secondary to an inhibition of fatty acid oxidation. We recently demonstrated that trimetazidine stimulates glucose oxidation in the heart second...
